چکیده

Although the enzyme chlorophyllase has been long known (13) only recently has evidence been presented for its probable function in chlorophyll a (chl a) biogenesis, namely, the demonstration that methyl chlorophyllide a (me chlide a) was transesterified with phytol in the presence of chlorophyllase yielding chl a (2). However, no evidence has been presented that wouild indicate the presence of me chlide a in green plants, a finding that would strongly support the above as the function of clhlorophyllase in chl a biosynthesis. This report describes the isolation and partial characterization of minor quantities of me chlide a from extracts of expanding wheat (Triticumiii aestivum) or passion flower leaves (Passiflora X alatocaerulea). This finding together with that described above suggests that the final steps in chl a biogenesis are

Pheophytin pheophorbide hydrolase (pheophytinase) is involved in chlorophyll breakdown during leaf senescence in Arabidopsis.

During leaf senescence, chlorophyll is removed from thylakoid membranes and converted in a multistep pathway to colorless breakdown products that are stored in vacuoles. Dephytylation, an early step of this pathway, increases water solubility of the breakdown products. It is widely accepted that chlorophyll is converted into pheophorbide via chlorophyllide. However, chlorophyllase, which conver...

Reexamination of chlorophyllase function implies its involvement in defense against chewing herbivores.

Chlorophyllase (CLH) is a common plant enzyme that catalyzes the hydrolysis of chlorophyll to form chlorophyllide, a more hydrophilic derivative. For more than a century, the biological role of CLH has been controversial, although this enzyme has been often considered to catalyze chlorophyll catabolism during stress-induced chlorophyll breakdown. In this study, we found that the absence of CLH ...

Chlorophyll Formation in Greening Bean Leaves during the Early Stages.

In the evolution of the absorption spectrum of etiolated bean leaves (Phaseolus vulgaris L.) following illumination, a rapid photoconversion of 50% or more of the active protochlorophyllide at room temperature is followed by a shift of the chlorophyll(ide) absorption maximum: C(678)--> -->C(684)-->C(672 nm). Kinetic studies at 2 C and the absence of an isosbestic point provide evidence for an i...

A role for chlorophyllide a oxygenase in the regulated import and stabilization of light-harvesting chlorophyll a/b proteins.

The Arabidopsis CAO gene encodes a 52-kDa protein with predicted localization in the plastid compartment. Here, we report that CAO is an intrinsic Rieske iron-sulfur protein of the plastid-envelope inner and thylakoid membranes. Activity measurements revealed that CAO catalyzes chlorophyllide a to chlorophyllide b conversion in vitro and that the enzyme was only slightly active with protochloro...

Melatonin Improves the Photosynthetic Apparatus in Pea Leaves Stressed by Paraquat via Chlorophyll Breakdown Regulation and Its Accelerated de novo Synthesis

The positive effect of melatonin on the function of the photosynthetic apparatus is known, but little is known about the specific mechanisms of melatonin's action in plants. The influence of melatonin on chlorophyll metabolism of 24-day-old Pisum sativum L. seedlings during paraquat (PQ)-induced oxidative stress was investigated in this study. Seeds were hydro-primed with water (H), 50 and 200 ...
